Abstract

Procede d'isolation d'un melange d'enzymes utile dans le traitement de tissus devitalises a partir de bromelaine brute. Le procede comprend les etapes suivantes: 1) suspension de la bromelaine brute dans une solution faiblement basique, de preference une solution de borate de sodium, pour dissoudre selectivement le melange d'enzymes actifs; 2) separation des solides non dissouts de la solution; et 3) extraction des petites molecules ayant un poids moleculaire de 10000 ou moins de la solution. Le produit possede une activite de 15 a 35 unites d'activite de protease par milligramme de proteines en presence de cysteine et de 0,5 a 5 unites d'activite de collagenase par milligramme de proteine. Le melange d'enzymes est dissout dans un solvant porteur inerte contenant au moins 60% d'eau avant application sur les tissus devitalises.
A process for isolating an enzyme mixture useful in the treatment of devitalized tissue from crude bromelain. The process comprises the steps of (1) suspending the crude bromelain in a weakly basic buffer, preferably sodium borate buffer, to selectively dissolve the active enzyme mixture; (2) separating the undissolved solids from the solution; and (3) removing small molecules having a molecular weight of 10,000 or less from the solution. The product exhibits 15 to 35 units of protease activity per milligram of protein in the presence of cysteine and from 0.5 to 5 units of collagenase activity per milligram of protein. The enzyme mixture is dissolved in an inert solvent carrier containing at least 60 percent water prior to application to the devitalized tissue.
